Abstract 

Background: Ulcerative colitis (UC) is a common and chronic gastrointestinal disorder characterized by intestinal inflammation and mucosal tissue damage. Reactive oxygen species (R03) play a pathogenic role in UC.
Objective: Investigate the anti-inflammatory and antioxidant effects ofKetotifen, a selective stabilizer ofmast celis andahistamineHi receptor antagonist,intreatmentthe acetic acid(A)inducedmodelforUlcerative colitis in rats.
Methods: Intrarectal instillation of acetic acid (Imi 5% v/v) was usedto induce UC. Ketotifen was used at dose of 2 mg/kg. Rats received treatment for 5 consecutive days. On day 6”, the rats were sacrificed, colon was removed, then the colon weightllength ratio, tissue glutathione (03H) concentration, macroscopic, and histopathological evaluations, ivere performed.
Resuits: The levels of colon weightllength ratio were decreased significantly (P < 0.05). The glutathione (03H) concentration vas increased significantly (P <0.05). The macroscopic and histopathological parameters were decreased, but it didn’t reduce significantly in the Ketotifen treated groups, comparedto the acetic acid-treated group.
Conclusions: The anti-inflammatory and anti oxidant effects of Ketotifen in treating the UC are due to its potential to reduce the colon weightllength ratio, increased 03H, and improving the macroscopic, and histological markers.
